Flavivirus Clickmers

Clickmers® are chemically modified DNA Aptamers designed for high specificity and affinity. The modifications, which are introduced to the ssDNA backbone using Click-Chemistry, provide opportunities for Clickmer-target-interactions that are impossible to achieve with naive DNA or RNA aptamers.

What are Flaviviruses?

Flaviviruses are a genus of viruses belonging to the Flaviviridae family. They are single-stranded, positive-sense RNA viruses that are primarily transmitted by arthropods, such as mosquitoes and ticks.

Flaviviruses are small enveloped RNA viruses causing severe diseases such as hemorrhagic fever and neurological complications in humans. Infections are increasing and emerging in new regions. Serological detection of infections is complicated due to cross-reactivity of the antibodies, making it hard to differentiate between flavivirus

Nucleic acid testing (NAT) methods face significant challenges due to the inherently low viral load present in samples, which can hinder accurate detection and quantification.

Upon flavivirus infection, hexameric NS1 is secreted from infected cells and can be detected in patients serum. Both in sequence as well as in structure, these NS1 proteins are highly homologous leading to cross-reactivity issues for monoclonal antibodies.

Flavivirus Clickmers Information

All Clickmers were selected and work best in "Buffer C" [Cat# 4101] and are available with a variety of different labels including but not limited to biotin, amine, Cy3, Cy5, and FAM. Clickmers are stored in buffer at pH 7.4.

Recommended storage conditions are: room temperature for 3 months, 4-6 °C for 6 months, -20 °C for 1 year. Fluorescently labeled Clickmers need to be stored in the dark to avoid fluorophore bleaching.

Flavivirus Clickmers Comparison

Figure 1. Clickmers against NS1 from TBEV, Yellow Fever and West Nile Virus show high specificity

Direct ELISA of Clickmers raised against various flavivirus NS1 proteins: The Clickmers show no cross-reactivity with NS1 proteins of the other, closely related to flaviviruses.

Figure 2. The pan-Dengue NS1 Clickmer recognised all four serotypes without other cross-reactivities

Direct ELISA of Clickmer raised against four serotypes of Dengue NS1: The Clickmers show no cross-reactivity with NS1 proteins of the other, closely related, flaviviruses.
